Tenofovir Alafenamide (TAF)
« on: February 19, 2014, 09:25:32 AM »
Does anybody have information on the progress of Gilead's tenofovir alafenamide (TAF)?  How close are we to actually seeing it on the market? And will it simply replace the current form of tenofovir in Truvada, Stribild, and Atripla or will there be three new drugs?

The patent on the existing drug doesn't expire until 2017 (or so)  so I wouldn't expect much before that.

IIRC as of now Gilead is only conducting phase 3 trials of this drug in combination with elvitegravir, cobicistat, and emtricitabine as a all-in-on-drug and NOT as a stand alone, which resulted in the following letter last year from Treatment Action Group:

... unless anyone has heard of a response and/or update to this. Yet another example of being jerked around needlessly by a drug company.

I've been enrolled in a TAF trial for about 7 months now.  In addition to taking Tivicay, I also take two Truvada pills.  One pill looks exactly like the Truvada I've always taken - a large blue tablet.  The other is also blue but it is about 1/3 the size of the original Truvada.  Obviously, one dosage is a sugar pill.  When the study concludes in about 18 months, I'm told that it will be revealed to me which formulaton I'm taking and I'll be able to choose between original Truvada and a new formulation using TAF.

In any case, for those of you who are concerned that Gilead won't issue a new stand-alone Truvada, it's quite clear that Gilead is intending to do that, perhaps alongside the old version.
