 :D a new drug outlook dealing with hiv genetic makeup cripples the virus to replicate. early stages don't know yet..anybody hear of it? or what stages... ???
mutagenesis sounds a bit like mutant x these days :D
intention to :cripple virus to replicate:// latent resevoirs or resting no idea?

    The extraordinarily high mutation rate of HIV suggests that the viral population exists near the threshold for viral viability. An increase in mutation could mediate a lethal increase in the already high proportion of defective viruses. We initially tested this hypothesis by culturing HIV-infected cells in mutagenic deoxyribonucleoside analogs. In the presence of 5-hydroxydeoxycytidine, there was a loss of viral replication after sequential passages of HIV in human CEM cells (Loeb LA, et al.: Proc Natl Acad Sci USA 1999;96:14921497). We now propose to use mutagenic ribonucleosides to induce lethal mutagenesis. The incorporation of mutagenic analogs into the HIV genome by the host cell RNA polymerase, rather than by the viral reverse transcriptase, augurs well for the avoidance of resistant viruses.

Hi again Puffy -

seems like they are giving providing a modified ribonucleosides to infected cells.  These are components necessary for RNA/DNA synthesis and replication of the viruses. 

The idea is that the modified nucleosides will be incporporated into replicating viral genome by the host cell's RNA polymerase during viral replication causing rapid rise in lethal mutations.  This polymerase is responsible for the incorporation of ribonuclesides into the new copies.

Since this incorporation would be mediated by the host cell (our) machinery (i.e. RNA polymerase) they contended that this would avoid viral resistance since it is outside the scope of the viral reverse transcriptase.

This study was done in 2000 and I have not yet done a search about the feasibility of such work in animals or humans since this was done in vitro.

My guess is that it has too many possibilites to cause back things for health cells (all type too..)

Ahhhhhhh fuck the ribosome in infected cells? Hmmm sounds dangerous, how does it select infected cells to start with, and there is a high risk of actually producing super-mutants, no?

I would think so.  Like I said thie research was done 7 years ago and I am not sure how much more has been done. I would think that this could have far reaching side effects as well as diffculty in delivery.

I don't know a lot about it, just what I read in the reference provided so I would have to do a little reading.  But, as I do puruse the literature a bit, I have not seen much recent news regarding this path for theraperutics.

yes, I'm sure I read that there were trials of something using this philosophy going on, just not sure if it's the same thing.  There was a thread on here about it somewhere. Anyone find it?
