Thanks, NYCguy.  This is an interesting chart with a few surprises.   Who knew that dendritic cells had been picked up by U. Pittsburgh?   The cells have gone from that French/Brazilian study to the Far East to Canada to Pennsylvania.  Conspicuously absent from the chart are the Ensoli and Emory U. vaccines (unless they're there under some different names).

DermaVir is still in phase I (well, I/II)?  It seems as though that's been around for ages...

I met someone in my support group who had a gene therapy at UCLA which sounds similar to the Johnson/Tibotec mentioned above and he was off meds for over a year and good results no real increase in VL


Well, if GMAT reduces viral load to undetectable and keeps it there... Even if it's not forever
Isn't that sign enough that it works?
This is a general comment, not specific to this particular therapy and research.

On the second quote from a different poster, "No" it's not a sign that it works. You are assuming a causal relationship between some treatment, like GMAT, and good results. "Controls" are designed to isolate factors, so that your optimistic statement could be proven to be true. Very difficult process.

I was just skimming here and noticed a similar emotional current in this serious exchange and the not so serious over the probable "Bob Beck" quackery promoted by a new member to our site. Human beings want solutions. Some folks want a Jim Jones messiah to "cure" them with the secret substance the government/pharmas/white people/rich/corrupt MDs won't give us. And smarter folks may want results published right away and meaningful results from tiny samples (5 folks left in some study group), but science doesn't work so easily. This strong patient might have been in the control group receiving nothing for all I know. He could have been me. 

I am not part of that UCLA study, and I have long hoped for good things from gene therapy. Guess what? I had several VL measures of undetectable and a couple more under 1000 during the second and third years off meds. Does that sound exciting? Well, now past 6 years of no meds, my VL is climbing slowly and probably won't ever dip down below 10-20k without meds. My CD4s slowly and fairly steadily are dropping about 300 decline over 6 years. So there's no miracle here, just a pretty good immune system doing its best. Without a large sample and other measures, we don't know if this treatment has had any positive effect. So, let's not jump on a bandwagon. Good research usually leads to other good research. But progress is slow. Optimism is great, but if I hear "cure for hiv" (not from you guys or these researchers, I mean in general) one more time I'm gonna wretch. Better treatments working in entirely different ways, that's probably our future. Successful gene therapy still seems far off.

As far as the Ensoli trial goes, remember, they really just got started late last year or early this year in the phase 2 trials.  I would expect to hear nothing until near the end of the year or next year.  As far as I see it, no news is good news... it means the trial hasn't completely failed and is still ongoing... at least I would assume.  Also, I believe the start of phase 2 was delayed as they scrambled to get public funding.  Realistically, I'm hoping to see a therapeutic vaccine come to market in about 5 - 10 years (and it probably won't work for everybody.. and maybe not forever either).  Anything sooner is an unexpected surprise.

Correct me if I am wrong, but obviously you are in some gene therapy experience and it sound like all the other meds that the virus start to mutate and resist against the "therapy" itself. right?

Sounds very depressing though :'( :'(

He is merely stating that he has had experience of decreased viral load without gene therapy...therefore a decreased viral load on its own may happen independently of any therapy simply due to natural immmune system fluctuations, and so cannot be used, on its own, as proof that any particular therapy works.

Regarding the trial results that were supposed to be released in Feb, and still missing.    Didn't someone in this forum say that they had corresponded with Michael Delane, the guy in the San Francisco trial, about his experience so far with the therapeutic vaccine.  He seemed very comfortable talking in the past.  Any way of contacting him now?

Exactly, lonbjny! Sorry, had not been watching this thread closely. Thanks for correcting the misunderstanding. Yeah, misinterpretations (by us, not scientists) of data was pretty much my point. Cause and effect is the issue, my issue.

...And to HAHAHA, I find my medical history to this day pretty heartening, hardly depressing.

And no gene therapy ever (Pretty rare anyway) and only one treatment (3TC + D4T, 1995-2000). I don't volunteer for treatment studies just because I watch them...Sort of like I never buy a new model car the first two years of production...too many kinks left.   That's my preferred take on "new therapies"8) -megasept

Hi, last year I heard reports about a gene therapy phase 1 or 2 trial (involving Johnson & Johnson) that was due to release results in Feb 07.

It has approx 70 recruits with 50% on placebo.

One guy went public with his results as he had achieved undetectable viral load.

Does anybody know the trial. And does anybody know anything about the due date for results?

well guys, I wrote to J&J, which owns Tibotec and after about a month, I actually got a response.  To recap, this is the Johnson/Tibotec study where they put your blood through a dialysis-type machine and permanently alter your white cells to make them hiv-resistant.  This is not the Virxsys study Appleboy is trying to get into, but rather the one the guy referred to in the above SF chronicle article was in.  So we wait again:

Unfortunately we experienced some delays in the implementation of this study and therefore it is still ongoing.  It is forecast that the last patient will complete last visit in Q1 2008, and the final results will be available Q2/3 2008.

What would happen to HAART-sales if this kind of therapy becomes the standard therapy? Maybe the big companies who already have a number of HAART-meds on the market, are not interested in this kind of development...

Hey All

There is a great article in a recent issue of Science on "Building an HIV-Proof Immune System." They list several current "gene therapy" clinical trials which are either going on now or about to start in the near future.

Unfortunately you can't easily get it online as you have to either subscribe to Science or be a member of AAAS.  And since I don't want to jeopardize my science career as it is just getting started I'm trying to avoid breaking any copy right laws.  At any rate this is the citation - you could get the article at the library if really interested.
